Catalog Description

Alirocumab is a recombinant, fully human IgG1 monoclonal antibody directed against PCSK9. It binds PCSK9 and inhibits PCSK9-mediated degradation of hepatic low-density lipoprotein receptors, thereby increasing LDL-receptor availability for clearance of LDL cholesterol. The molecule is produced in Chinese hamster ovary cell suspension culture and has an approximate molecular weight of 146 kDa.

Catalog descriptions reflect research context only. Not a medical claim or treatment recommendation.

Research Profile

Half-Life
Median apparent steady-state half-life: 17–20 days with 75 mg or 150 mg administered subcutaneously every 2 weeks; approximately 12 days when administered with a statin, according to the FDA labeling.
Onset
Following a single subcutaneous administration of 75 or 150 mg, maximal suppression of free PCSK9 occurred within 4–8 hours. Median time to maximum serum concentration after 75–300 mg subcutaneous administration was 3–7 days. In a healthy-subject pharmacokinetic study, free PCSK9 was reduced to approximately zero between study days 3 and 4.
Administration Routes (Research)
subcutaneous

Reported Research Effects

Reflects preclinical/research literature context only. Not a medical claim, dosing instruction, or treatment recommendation.

Research Notes

Regulatory product information documents subcutaneous regimens including 75 mg or 150 mg every 2 weeks and 300 mg every 4 weeks in specified clinical contexts. A randomized pharmacokinetic study evaluated a single 75 mg subcutaneous administration using a pre-filled pen at the abdomen, upper arm, or thigh; systemic exposure was similar across injection sites. These regimen and administration details are recorded as neutral study/reference information and are not individualized dosing instructions.

Supplementary research context only. Not medical advice or a treatment recommendation.

Source-Backed Observations

Alirocumab consists of two disulfide-linked human heavy chains, each linked to a human kappa light chain, with one N-linked glycosylation site in each heavy-chain CH2 domain. It is supplied as a sterile, preservative-free, clear, colorless to pale-yellow solution at approximately pH 6.0.
