Catalog Description

Resmetirom is a synthetic small molecule that selectively activates thyroid hormone receptor-beta, the predominant thyroid hormone receptor isoform in the liver. Its molecular formula is C17H12Cl2N6O4 and its molecular weight is 435.22 g/mol. The compound is indicated in the United States, under the name Rezdiffra, for adults with noncirrhotic metabolic dysfunction-associated steatohepatitis with moderate to advanced liver fibrosis, in conjunction with diet and exercise.

Research Notes

The current DailyMed label was updated in July 2026. U.S. approval was granted on March 14, 2024, under the accelerated-approval pathway for noncirrhotic MASH with moderate to advanced fibrosis. The cited sources describe tablet strengths of 60 mg, 80 mg, and 100 mg and oral administration; they do not establish a general research-material presentation or noncommercial reference size.

Source-Backed Observations

Resmetirom is a partial agonist of thyroid hormone receptor-beta. In an in vitro functional assay, it produced 83.8% of the maximum response of triiodothyronine at THR-beta, with an EC50 of 0.21 micromolar. THR-beta activation in the liver is associated with reduced intrahepatic triglycerides. Resmetirom is metabolized by CYP2C8; its major metabolite is MGL-3623. It is greater than 99% protein-bound and is also an inhibitor of CYP2C8, OATP1B1, OATP1B3, and BCRP according to the current U.S. prescribing information.
