Best-Effort Research Profile

DLin-MC3-DMA, commonly abbreviated MC3, is an ionizable amino lipid containing two linoleyl-derived unsaturated hydrocarbon chains and a dimethylaminobutanoate head group. It is used in lipid nanoparticles for delivery of siRNA and other nucleic-acid cargos and is the ionizable lipid component of the approved patisiran lipid complex formulation.

Source review pending. This descriptive profile is shown from the available source material as a best-effort educational reference. It has not passed the full verification bar and is not a dosing instruction, medical claim, or treatment recommendation.

Research Notes

DLin-MC3-DMA was identified through optimization of ionizable lipids for hepatic siRNA delivery. The 2012 Jayaraman study reported an optimized lipid nanoparticle formulation containing 50 mol% DLin-MC3-DMA and described an siRNA ED50 of approximately 0.005 mg/kg in mice and less than 0.03 mg/kg in cynomolgus monkeys; these are study-level siRNA payload doses, not standalone DLin-MC3-DMA doses. The same study associated high delivery activity with an ionizable-lipid pKa range of approximately 6.2-6.5 and reported a pKa of approximately 6.44 for MC3. The common MC3 lipid nanoparticle composition is DLin-MC3-DMA/DSPC/cholesterol/PEG-lipid at approximately 50:10:38.5:1.5 mol%, although composition varies by study. The current U.S. product labeling for ONPATTRO lists 13.0 mg/mL DLin-MC3-DMA in the lipid complex. No standalone compound half-life, onset, solubility, or storage specification was established from the authoritative sources reviewed.
