Best-Effort Research Profile

7-Hydroxymitragynine is a stereochemically defined alkaloid with molecular formula C23H30N2O5 and molecular weight approximately 414.5 g/mol. It occurs naturally as a minor constituent of Mitragyna speciosa and is also formed as an oxidative metabolite of mitragynine in biological systems. Pharmacology studies describe activity at opioid receptors, with predominant mu-opioid receptor agonism and substantially greater potency than mitragynine in several experimental assays. No FDA-approved drug containing 7-hydroxymitragynine has been identified.

Source review pending. This descriptive profile is shown from the available source material as a best-effort educational reference. It has not passed the full verification bar and is not a dosing instruction, medical claim, or treatment recommendation.

Research Profile

Half-Life
Human pharmacokinetic assessment after oral mitragynine administration reported a 7-hydroxymitragynine apparent half-life of approximately 1.7-4.7 hours; a validated human half-life after direct administration of isolated 7-hydroxymitragynine has not been established. In male Sprague-Dawley rats, direct oral and intravenous pharmacokinetic studies have been reported, but the cited abstract does not provide a single elimination half-life value.
Onset
Not established as a reliable human clinical parameter. In mice, antinociceptive activity was assessed after oral or subcutaneous administration in preclinical time-course studies.
Administration Routes (Research)
oralsubcutaneousintravenous

Reported Research Effects

Research Notes

In the original mouse antinociception study, subcutaneous doses of 2.5-10 mg/kg and oral doses of 5-10 mg/kg were evaluated. A later rat pharmacokinetic study evaluated direct oral administration at 5 mg/kg and intravenous administration; the study reported rapid absorption, low oral bioavailability of approximately 2.7%, oral Tmax approximately 0.3 hours, intravenous clearance approximately 4.0 L/h/kg, and volume of distribution approximately 2.7 L/kg. These are animal-study reference parameters and are not human dosing recommendations.

Supplementary research context only. Not medical advice or a treatment recommendation.

Source-Backed Observations

Natural kratom leaf generally contains 7-hydroxymitragynine at trace or minor levels, substantially below the concentrations reported for enhanced or concentrated preparations. Human evidence is limited and largely concerns 7-hydroxymitragynine measured as a metabolite after mitragynine or kratom administration rather than direct administration of isolated compound. FDA states that 7-hydroxymitragynine is not approved for medical use.
